Author information: (1)Division of Basic Science, Research Institute, National Cancer Center, Goyang, Republic of Korea. kimsooyoul@gmail.com Many reports have shown that the expression of transglutaminase 2 (TG 2) is increased in inflammatory diseases. Transglutaminase 2 (TG2) is the most widely distributed and most abundantly expressed member of the transglutaminase family of enzymes, a group of intracellular and extracellular proteins that catalyze the Ca 2+ -dependent posttranslational modification of proteins. Tranglutaminase-2 (also known as TGM2, TG2), is a widely expressed, multifunctional protein. One protein is an enzyme called tissue transglutaminase, or TTG, that is found in many cells of our body.

TG2 crosslinks proteins between the ε-amino group of a lysine residue and the γ-carboxamide group of glutamine residue, creating an inter- or intramolecular bond that is highly resistant to proteolysis (protein degradation). Transglutaminase 2 antibody [N3C3] detects Transglutaminase 2 protein at cytoplasm by immunofluorescent analysis.

PMID: 22386570 Schizophr Res. maj 2010, 118 (1-3):240-7.doi:  TG2 is a multifunctional enzyme that belongs to transglutaminases which catalyze the crosslinking of proteins by epsilon- (gamma-glutamyl)lysine isopeptide bonds. Similarly to other transglutaminases, tTG consists of a GTP/ GDP binding site, a catalytic domain, two beta barrel and a beta-sandwich. Transglutaminase 2 (TG2) is an inducible transamidating acyltransferase that catalyzes Ca 2+ -dependent protein modifications. It acts as a G protein in transmembrane signalling and as a cell surface adhesion mediator, this distinguishes it from other members of the transglutaminase family. Transglutaminases are enzymes that in nature primarily catalyze the formation of an isopeptide bond between γ- carboxamide groups (- (C=O)NH 2) of glutamine residue side chains and the ε- amino groups (-NH 2) of lysine residue side chains with subsequent release of ammonia (NH 3).

TG2 crosslinks proteins between the ε-amino group of a lysine residue and the γ-carboxamide group of glutamine residue, creating an inter- or intramolecular bond that is highly resistant to proteolysis (protein degradation). Transglutaminase 2 (TG2) is the most widely distributed member of the transglutaminase family with almost all cell types in the body expressing TG2 to varying extents. In addition to being widely expressed, TG2 is an extremely versatile protein exhibiting transamidating, protein disulphide isomerase and guanine and adenine nucleotide binding and hydrolyzing activities.
